Bacterial Cell Envelope Lysis and Hemotoxicity of Peptides Previously isolated From African Catfish, Clarias gariepinus

چکیده

Background: The skin mucus layer of fish is endowed with biologics including, Antimicrobial peptides (AMPs) that offer a first line defence against pathogens. Such can either inhibit bacterial growth or completely kill the bacteria and hence are regarded as viable alternative to traditional antibiotics, in addressing ever-increasing incidences antimicrobial resistance. However, one major hurdles AMPs use their poor haemolytic profile. As result, thorough evaluation prospective AMPs’ cell membrane disruption hemolytic potentials early phases drug discovery critical. current study presented destruction well hemo-compatibility previously isolated from African catfish, Clarias gariepinus. Methods: A peptide C. gariepinus were profiled using 15% Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE). electrical conductivity alkaline phosphatase assays utilised measure envelope lysis activity classical mode action peptides. Afterwards, fresh Rabbit blood cells then for vitro assay. Results: found be about 5 kDa molecular weight with, ability damage causing significant leakage periplasmic enzyme cytoplasmic electrolytes. Even at highest extract concentration 100 μg/mL, no hemolysis was observed on rabbit [3.63%;P>.05], signifying safety normal mammalian cells. Conclusion: findings this pointed out potentially safe source leads; however, further studies still required search possibly maximum dose host but effective infecting bacteria.

Histopathological Effects of Glyphosate on Juvenile African Catfish (Clarias gariepinus)

Most aquatic herbicides have undergone some toxicity testing for effects on non-target aquatic organisms, little of this testing has been conducted on early life stages of Clarias gariepinus. African catfish C. gariepinus were exposed to acute concentrations of glyphosate for 96hrs. The lethal concentration (LC ) 5O value of glyphosate was 0.295 mg l for 96h of exposure. The mean mortality perc...

Responses of growth rates and growth hormone levels of African catfish (Clarias gariepinus) to stocking density

In order to estimate responses of growth rate and growth hormone of Clarias gariepinus to stocking density, fish (initial weight 30.71 ± 0.89 g) were stocked in 120-L tanks at densities of 35, 65, 95, and 125 kg/m3 for 60 days. On day 30 and 60, the fish growth rates were measured, pituitary growth hormone (GH) mRNA levels and serum GH concentrations were examined by real-time qPCR and ELISA te...

Occurrence and antibiotic susceptibility of fish bacteria isolated from Oreochromis niloticus (Nile tilapia) and Clarias gariepinus (African catfish) in Uganda

The intention of this study was to identify the bacterial pathogens infecting Oreochromis niloticus (Nile tilapia) and Clarias gariepinus (African catfish), and to establish the antibiotic susceptibility of fish bacteria in Uganda. A total of 288 fish samples from 40 fish farms (ponds, cages, and tanks) and 8 wild water sites were aseptically collected and bacteria isolated from the head kidney...

Induction of triploidy with caffeine treatment in the African catfish (Clarias gariepinus)

Induction of triploidy is one of the biotechnological methods in aquaculture used for genetic manipulation. It refers to a state where organisms have three complete sets of chromosomes instead of two and can result in sterility. Caffeine treatment that is safe and inexpensive, serve to induce triploidy in catfish. To suppress the second meiotic division, fertilized eggs were exposed to three di...
